Immunohistochemistry localises myosin-7a to cochlear efferent boutons

چکیده
Background: Myosin 7a is an actin-binding motor protein involved in the formation of hair-cell stereocilia both cochlea and vestibular system. Mutations myosin are linked to congenital hearing loss present 50% Type-1 Usher syndrome patients who suffer from progressive system dysfunction. Methods: often used visualise sensory hair cells due its well characterised localised expression profile. We thus conducted myosin-7a immunostaining across all three turns adult rat organ Corti cells. Results: As expected, we observed staining inner outer cells. Unexpectedly, also strong medial olivocochlear efferent synaptic boutons contacting Efferent bouton was cochlea. verified this localisation by co-staining with a known marker, vesicular acetylcholine transporter. Conclusions: In addition role maintenance, or certain variants might play transmission ultimately influence cochlear gain regulation. Our immunohistochemistry results should be validated other methods confirm these serendipitous findings.

Regulation of myosin 5a and myosin 7a.
The myosin superfamily is diverse in its structure, kinetic mechanisms and cellular function. The enzymatic activities of most myosins are regulated by some means such as Ca2+ ion binding, phosphorylation or binding of other proteins. In the present review, we discuss the structural basis for the regulation of mammalian myosin 5a and Drosophila myosin 7a. We show that, although both myosins hav...
متن کاملEfferent feedback slows cochlear aging.
The inner ear receives two types of efferent feedback from the brainstem: one pathway provides gain control on outer hair cells' contribution to cochlear amplification, and the other modulates the excitability of the cochlear nerve. Although efferent feedback can protect hair cells from acoustic injury and thereby minimize noise-induced permanent threshold shifts, most prior studies focused on ...

متن کاملA FERM domain autoregulates Drosophila myosin 7a activity.
Full-length Drosophila myosin 7a (myosin 7a-FL) has a complex tail containing a short predicted coiled coil followed by a MyTH4-FERM domain, an SH3 domain, and a C-terminal MyTH4-FERM domain. Myosin 7a-FL expressed in Sf9 cells is monomeric despite the predicted coiled coil. We showed previously that Subfragment-1 (S1) from this myosin has MgATPase of V(max) approximately 1 s(-1) and K(ATPase) ...
متن کاملRegeneration of cochlear efferent nerve terminals after gentamycin damage.
Chickens recover auditory function after hair cell loss caused by ototoxic drug damage or acoustic overstimulation, indicating that mechanisms exist to reestablish appropriate neuronal connections to regenerated hair cells. However, despite similar hair cell regeneration times, hearing recovery takes substantially longer after aminoglycoside than after sound damage. We have therefore begun exam...
